How the major stock indexes fared Thursday

It's only been a day, but November on Wall Street is already looking a lot better than October.

Strong economic data and corporate news converged Thursday to give U.S. stocks their best day since mid-September. Positive signs about the job market and higher auto and retail sales reports pushed stock futures up before the market opened. The Dow Jones industrial average rose 100 points the first half hour of trading. At 10, two more strong reports came out and pushed the Dow up as much as 177 points. It fell back some, but held a steady gain for the rest of the day.

The Dow Jones industrial average rose 136.16 points, or 1 percent, to close at 13,232.62.

The Standard & Poor's 500 rose 15.43 points, or 1.1 percent, to 1,427.59.

The Nasdaq composite index rose added 42.83, or 1.4 percent, to 3,020.06.

For the week:

The Dow is up 125.41 points, or 1 percent.

The S&P 500 is up 15.65 points, or 1.1 percent.

The Nasdaq is up 32.11 points, or 1.1 percent.

For the year:

The Dow is up 1,015.60 points, or 8.3 percent.

NEW YORK (AP) ? The vast destruction wreaked by the storm surge in New York could have been prevented with a sea barrier of the type that protects major cities in Europe, some scientists and engineers say. The multibillion-dollar price tag of such a project has been a hindrance, but may appear more palatable after the damage from Superstorm Sandy has been tallied.

"The time has come. The city is finally going to have to face this," said oceanography professor Malcolm J. Bowman at Long Island's Stony Brook University. He has warned for years of the potential for a catastrophic storm surge in New York and has advocated for a barrier.

Invented by Bowman and his colleague Douglas Hill, two European engineering firms have drawn up proposals for walling most of New York off from the sea, at a price just above $6 billion.

Before the storm, Mayor Michael Bloomberg's administration had said it was working to analyze natural risks and the effectiveness of various coast-protection techniques, including storm-surge barriers. But officials had noted that barriers were only one of many ideas, and they have often emphasized more modest, immediate steps the city has taken, such as installing floodgates at sewage plants and raising the ground level while redeveloping a low-lying area in Queens.

"It's a series of small interventions that cumulatively, over time, will take us to a more natural system" to deal with climate change and rising sea levels, Carter H. Strickland, the city's environmental commissioner, told The New York Times this summer.

Engineers know this approach as "resilience" ? essentially, toughening the city piece by piece to make it soak up a surge without major damage. But the European engineering firms whose barriers protect the Netherlands and the Russian metropolis of St. Petersburg see this as unrealistic, given the vast amount of expensive infrastructure that underpins New York.

"How does New York as a city retreat into resilient mode? It's just difficult to see how that would happen," said Graeme Forsyth, an engineer for CH2M Hill in Glasgow, Scotland.

Sandy sent a record 14-foot storm surge into New York Harbor, flooding subway tunnels and airports. It forced the closure of the stock market for two days, the first time that's happened for weather-related reasons since 1888. There's no estimate yet for the cost of the devastation in New York City, but forecasting firm IHS Global Insight put the cost of the damage along the coast at $20 billion, plus $10 billion to $30 billion in lost business.

Forsyth has worked on St. Petersburg's barrier, which consists of 16 miles of levees and gates shielding the city, built on what was once a swamp, from the Baltic Sea and the river Neva. The centerpiece of his firm's early-stage proposal for New York is a levee-like barrier that would stretch five miles from the Rockaway peninsula in Queens on Long Island to the Sandy Hook promontory in New Jersey. The barrier would stop a surge of 30 feet, twice the height from Sandy. Gaps would allow ships, river water and tides through, but movable gates could close off all of New York Bay from the Atlantic when necessary. The barrier would protect most of the city, with the exception of Rockaway itself. It would also shield parts of New Jersey.

To be sure, some scientists have reservations about the storm-surge barrier concept.

Some are concerned about how the structures could affect tidal flow and other environmental features of New York Harbor ? and about whether barriers would be socially fair.

"Who gets included to be behind the gate, and who doesn't get included? ... How do you make that decision in a fair way?" Robert Swanson, an oceanographer who is Bowman's colleague at Stony Brook, said in an August interview.

Other experts question whether barriers would even work in the long term. Klaus H. Jacob, a Columbia University climate-risk researcher who has advised New York City officials, has noted that given the unknowns of climate change, any system designed now could prove inadequate in the future.

But advocates believe New York needs to take bigger steps given its concentration of people and financial infrastructure.

"With the kind of protection that has been considered so far, you cannot protect a multimillion-inhabitant city that runs part of the world economy," said Piet Dircke, who has worked on the extensive system of sea barriers in the Netherlands with the Dutch engineering firm Arcadis.

His firm's proposal is to build a barrier in the Verrazano Narrows between Brooklyn and Staten Island, shielding Upper New York Bay. It would be supplemented by two smaller barriers, one between Staten Island and New Jersey and the other on the East River. Such a barrier would have protected Manhattan and much of Brooklyn and Staten Island from Sandy, but left southern Brooklyn and Kennedy Airport exposed.

Robert Trentlyon, a New York community activist who has been advocating for storm-surge barriers, sees the one-two punch of Hurricane Irene in 2011 ? which came within a foot of flooding subway stations in southern Manhattan ? and Sandy as a sign that the time has come.

"Having had two storm surges within one year, and their both being major ones, I just find it very difficult to think the city could not go ahead and act," the retired local newspaper publisher said by phone Sunday from his Manhattan apartment, which was left without power. His Chelsea neighborhood, though not his building, was among those that flooded.

In August, U.S. Rep. Jerry Nadler urged city officials to take a comprehensive look at storm-surge barriers, bulkheads and other flood-fighting devices.

After the storm, reactions from the government have been mixed, as the region battles to recover from the storm rather than looking at how to prevent the next disaster.

"We cannot build a big barrier reef off the shore to stop the waves from coming in," Bloomberg said Monday. But Gov. Andrew Cuomo opened the door to new ideas Tuesday, saying that the government has a responsibility to think about new designs and techniques to protect the city in the face of what look like increasingly frequent megastorms.

One doesn't have to go to Europe or New Orleans to find examples of massive sea barriers: The city of Providence, R.I., has been protected by a 3,000-foot gated barrier since 1966. Construction was prompted by two devastating hurricanes in 1938 and 1954. The barrier has prevented flooding of the low-lying parts of the city several times since then, including during Sandy.

"This is not far-out science or engineering," Bowman said. "This is easy to do."

"Easy" doesn't mean it would be something that could be put in place quickly. Even after politicians line up behind the project, funding, permitting and environmental studies are likely to take years.

"It could take 20 years before people even start pouring concrete," Bowman said.

ScienceDaily (Oct. 31, 2012) ? Scientists have discovered that breaking a biological signaling system in an embryo allows them to change the destiny of a cell. The findings could lead to new ways of making replacement organs.

The discovery was made in the laboratory of Joel H. Rothman, a professor in the Department of Molecular, Cellular, and Developmental Biology at UC Santa Barbara. The studies were reported in the interdisciplinary journal Genes and Development, and were carried out by Ph.D student Nareg Djabrayan, in collaboration with Rothman and two other members of the laboratory, Ph.D student Erica Sommermann and postdoctoral fellow Nathaniel Dudley.

"At some point along the way toward becoming part of a complete individual, cells become destined to choose a particular identity and long-term profession," Rothman noted. "Once a cell chooses who it will be, it locks onto that identity for the remainder of its life."

A cell that is destined to become a heart cell functions exclusively in the heart until it dies, and never chooses later to change jobs by becoming, for example, a brain cell. "If Oz's wizard possessed the powers he claimed, and had a spare brain lying around, he could switch it to a heart as a gift for the Tin Man. And he could reverse the trick for the Scarecrow," Rothman said.

Similarly, the researchers have found a way to unlock cells' destinies and lead them to take on a new profession.

The scientists found that a widely used cell signaling system, known as "Notch" signaling, causes cells to commit to a particular occupation, such as a skin or brain cell. When they blocked the signal by genetic manipulation, the researchers discovered that they could force a cell to change its destiny, such that they instead became cells of the intestine.

"We found that we could break the signal in such a way that cells would follow their usual destinies, but were somehow less committed to doing so: We could convince them to change professions long after they would normally refuse to do so," Rothman said.

The scientists made the discovery by harnessing the genetics of a tiny nematode worm known as C. elegans, a model animal that has become famed in fundamental studies in biomedicine, and has been the subject of six Nobel prizes.

The researchers discovered that shutting off the Notch signal in early embryos made it possible for them to change the destiny of cells much later on, at a stage when they normally could not. "The later cells seemed to remember what had happened to their great-grandparent cells," Rothman said. "Imagine if the experience of your great-grandparents predestined you to become a lawyer, dentist, or coal miner, and you could not choose to change your professional destiny. That is what is happening to cells whose cellular ancestors received the Notch signal."

The research was supported by the National Institute of Child Health and Human Development and the California Institute of Regenerative Medicine.

The discovery could someday help scientists develop new ways to produce tissues and organs in the laboratory that could be used to replace a patient's injured, diseased, or aged organs. By unlocking a cell's normal destiny, it may be possible to change it into an altogether different type of cell that could be used to grow a new organ for a patient -- or perhaps, a Tin Man.

First effective treatment of tumors arising from common genetic disease NF1, suggested by study

ScienceDaily (Nov. 1, 2012) ? Physician-researchers at Indiana University School of Medicine have reported the first effective therapy for a class of previously untreatable and potentially life-threatening tumors often found in children.

Announcing their findings in the online first edition of Lancet Oncology, the researchers said the drug imatinib mesylate, marketed as Gleevec as a treatment for chronic myeloid leukemia, provided relief to a significant number of patients with plexiform neurofibromas, tumors caused by neurofibromatosis type 1, or NF1.

"Although this was a small study, the results were significant, particularly given that such patients have had few treatment options for what can be a very debilitating disease," said first author Kent Robertson, M.D., Ph.D., associate professor of pediatrics. "We believe these findings warrant larger trials of both imatinib mesylate as well as other similar compounds that would appear promising in laboratory tests."

Affecting about 1 in every 3,000 children born, NF1 is the most common neurological disorder caused by mutations in a single gene. Including a much rarer related type, NF2, neurofibromatosis is a genetic disorder that is more prevalent than cystic fibrosis, Duchenne muscular dystrophy and Huntington's disease combined, according to the Children's Tumor Foundation.

The mutation produces a variety of symptoms, from mild to severe. Patients can develop caf? au lait spots and disfiguring tumors on or just under the skin. Internally, tumors can develop along nerve tissue and cause problems if they begin to press against vital organs or the windpipe. Some patients suffer from chronic pain.

The tumors have been nearly impossible to treat effectively. Because they are relatively slow growing, they are not affected by radiation treatments or cancer chemotherapy drugs. They also are often not good candidates for surgery because they are dangerously close to vital organs.

In the study reported in Lancet Oncology, of 23 patients who received the drug for at least six months, six experienced a 20 percent or more decrease in the volume of one or more plexiform neurofibromas, and 30 percent of patients had improvements in symptoms.

Primary investigator D. Wade Clapp, M.D., Richard L. Schreiner Professor and chairman of the Department of Pediatrics, noted that even relatively small reductions in tumor size can result in significant relief of symptoms for patients, such as improved breathing and restoration of bladder control.

In earlier laboratory research, the researchers determined that Gleevec was effective in tissue culture and mouse models of NF1 tumors after discovering that a cellular signaling mechanism that Gleevec targets in chronic myeloid leukemia also played an important role in development of NF1 tumors. Reporting that finding in the journal Cell in 2008, the research team also reported that in a compassionate use protocol, they had treated a 3-year-old girl with a life-threatening tumor compressing her airway. The girl's tumor shrank by half within three months of treatment.

Although Gleevec has been widely used as a treatment for chronic myeloid leukemia and has been prescribed in some cases for long periods without serious side effects, 13 of the initial 36 patients enrolled in the IU study dropped out before their results could be analyzed at six months of treatment. Nine of the 13 left due to problems taking the drug or side effects. The authors noted that they used the previously established maximum tolerated dose for Gleevec for this study to ensure that any drug activity against any NF1 tumors would be observed. This approach was taken among patients who had been living with slow-growing tumors for long periods of time and therefore were less likely to accept drug side effects than patients with malignant tumors, the authors suggested. Dosages have been modified in subsequent studies.
